| Background | The extracellular signal-regulated kinase 2 (ERK2, 42kDa), also known as mitogen-activated protein kinase 1 (MAPK1), is located in an integration point of the mitogen-activated protein kinase (MAPK) cascading pathways and have been implicated in the regulation of diverse cellular processes such as proliferation, differentiation, transcription regulation and development (1-4). Activation and inactivation of ERK2, a serine/threonine protein kinase, are tightly regulated by an upstream protein kinase MEK1/2 and dual-specificity protein phosphatases (DSUP). In human cancers and human disorders, aberrantly upregulated MEK1 activates its unique substrate ERK1/2. Therefore, the Ras-Raf-MEK1-ERK2 pathway has become an attractive target for anticancer therapeutics (5). |
