The activating transcription factor 2 (ATF2), known as CRE-BP1 (cAMP-responsive element binding protein 1), is a member of the leucine zipper family of DNA binding proteins and binds to both AP-1 and CRE DNA response elements of many viral and cellular promoters (1). ATF2 is activated by diverse cellular stresses such as inflammatory cytokines, genotoxic agents and UV irradiation. It is activated through phosphorylation at two phospho sites of Thr69 and Thr71 by upstream activators like JNK/p38 (MAPK14, SAPK2A) (2-4). GST-ATF2 [19-96] is preferentially used as a useful substrate for MAPK14 and was assayed using MAPK14 (P5049 and R5048) by luminescence-based ATP depletion assay.
Recombinant GST-ATF2 [19-96] is isolated from an E coli strain that carries the coding sequence of the human ATF2 [19-96] under the control of a T7 promoter. GST-ATF2 [19-96] can be applied in vitro phosphorylation assay, DNA and protein-protein interactions assays. For Research Purposes Only. The purified recombinant protein is greater than 95% homogeneous. 20 units are sufficient for a gel-mobility shift assay and 100 units are sufficient for a protein-protein interactions assay. 2 mg/ml (in 1X dilution buffer A) 1X dilution buffer A: 20 mM Tris-Cl (pH 8.0), 20% Glycerol, 100 mM KCl, 1 mM DTT and 0.2 mM EDTA.
