The protein catalyzes the phosphorylation of phosphatidylinositol-4-phosphate on the fifth hydroxyl of the myo-inositol ring to form phosphatidylinositol-4,5-bisphosphate. The gene is a member of the phosphatidylinositol-4-phosphate 5-kinase family. The encoded protein sequence does not show similarity to other kinases, but the protein does exhibit kinase activity. Additionally, the encoded protein interacts with p55 TNF receptor. Two transcript variants encoding different isoforms have been found for this gene. Pip5k2beta, is amplified in a subset of breast cancer cell lines and primary breast cancer samples that carry HER-2/Neu amplification. (1) It has been implicated that Pip4k2beta has a role in the development and/or progression of breast cancer.
The PIP5Kβ was expressed in Baculovirus system and purified by an affinity column in combination with FPLC chromatography. 1ug of PIP5kβK is sufficient to phosphorylate 1 ug of heat-inactivated nuclear or cytoplasmic extract at 30 C for 30 mins. The purified recombinant protein is greater than 95% homogeneous and contains no detectable protease, DNase and RNase activity 1 unit equals 1 nanogram of purified protein. 0.5 mg/ml (in 1X dilution buffer A) Buffer A: Tris-Cl, Glycerol, KCl, DTT and EDTA.
