Production of interleukin-1 and tumor necrosis factor (TNF) are inhibited by compounds called CSAIDs (cytokine-suppressive antiinflammatory drugs). 2 CSBPs (CSAID-binding proteins) have been characterized as molecular targets of pyridinyl-imidazole cytokine inhibitors. The 2 closely related targets are mitogen-activated protein kinases (MAPKs) CSBP1 and CSBP2. (1) MAPK14 is activated in response to many cellular stresses and also regulates the differentiation and/or survival of various cell types in vitro, including skeletal muscle cells and cardiomyocytes. Studies indicate that MAPK14 is required for placental organogenesis but is not essential for other aspects of mammalian embryonic development. (2)The inhibition of p38alpha MAPK blocks the release of vascular endothelial growth factor and suppresses tumor-promoted endothelial cell migration, a key step in angiogenesis.(3)
The MAPK14 was expressed in a Bacterial system and purified by an affinity column in combination with FPLC chromatography.
May be used in phosphorylation experiments.
Purified protein is greater than 95% homogeneous and contains no detectable proteases, DNase and RNase activity.
1 unit equals 1 nanogram of purified protein.
