 Purification and Quality Control  The Flag-tag recombinant protein is purified by affinity chromatography in combination with FPLC columns.  The purified NcoR1 is greater than 90% homogeneous based on SDS-PAGE analysis.
 Unit Definition (Activity) 1 unit equals 1 nanogram of purified protein.
 Applications Recombinant N-CoR can be used 1) for in vitro function studies including transcription; 2) for protein-protein interaction assay; and 3) for cell growth assay.
 Formulation and Storage The protein is in 20mM Tris-HCl pH7.9,100mM NaCl, 0.2mM EDTA, 1mM DTT and 20% glycerol. Stored at -70°C before use. Avoid repeated freeze thaw cycles.

 Background This gene encodes a protein that mediates ligand-independent transcription repression of thyroid-hormone and retinoic-acid receptors by promoting chromatin condensation and preventing access of the transcription machinery. It is part of a complex which also includes histone deacetylases and transcriptional regulators similar to the yeast protein Sin3p. This gene is located between the Charcot-Marie-Tooth and Smith-Magenis syndrome critical regions on chromosome 17. An alternatively spliced transcript variant has been described, but its full length sequence has not been determined.
