This gene encodes a protein that mediates ligand-independent transcription repression of thyroid-hormone and retinoic-acid receptors by promoting chromatin condensation and preventing access of the transcription machinery. It is part of a complex which also includes histone deacetylases and transcriptional regulators similar to the yeast protein Sin3p. This
gene is located between the Charcot-Marie-Tooth and Smith-Magenis syndrome critical regions on chromosome 17. An alternatively spliced transcript variant has been described, but its full length sequence has not been determined. Recombinant His tagged N-CoR protein was expressed in Baculovirus system and purified by an affinity column in combination with FPLC chromatography. Recombinant N-CoR can be used 1) for in vitro function studies including transcription; 2) for protein-protein interaction assay; and 3) for cell growth assay. The purified recombinant proteinis greater than 95% homogeneous based on SDS-PAGE gel analysis. N-CoR has been tested for deacetylation activity in vitro in the presence of HDAC3(Cat. #P1091). 1 unit equals 1 nanogram of purified protein. 0.4 mg/ml (in 1X dilution buffer A) Buffer A: Tris-Cl, Glycerol, KCl, DTT, EDTA.
